Research directions:
1. Study on the impact of metabolic pathways on the homeostasis and disease mechanisms of the heart and brain. The functional homeostasis of the heart and blood vessels is closely related to their metabolic state. Metabolic pathways are widely involved in the occurrence and development of cardiovascular and cerebrovascular diseases (such as aneurysms, myocardial infarction, stroke, etc.). Metabolic regulation is an important research field following gene regulation and protein regulation. The team has discovered that multiple metabolic molecules and pathways are involved in new pathophysiological mechanisms of cardiovascular diseases and their key target proteins, including macrophages activating inflammatory responses by releasing succinate; cell junction protein Cx43 maintaining the homeostasis of the cerebral vascular endothelial barrier through regulating NAD+ levels; smooth muscle cells mediating the release of putrescine and phenotypic transformation under aneurysm conditions; lipid metabolism pathways such as apo(a) and lipid lipoprotein signaling pathways affecting the new mechanisms of the cardiovascular system, etc.
2. Discover new active factors from endogenous and gut microbiota sources and their metabolic regulatory effects. In samples from patients with cardiovascular and cerebrovascular diseases, we have identified multiple active factors through multi-omics methods, and discovered their effects and mechanisms on metabolic-related diseases such as diabetes, fatty liver, and tumors. Among them, we have confirmed that the endogenous factor annexin A1 plays a role in different cell types in aneurysm, diabetic nephropathy, and obesity and exerts protective effects, and vascular endothelial metabolism regulates angiogenesis and tumor progression in tumors. The gut microbiota is another important source of human metabolism. The metabolic spectrum changes caused by the disorder of the gut microbiota during the disease process further affect the disease progression, thereby generating new theoretical systems such as the gut-heart axis, gut-brain axis, and gut-liver axis. The team has first identified the new gut microbiota metabolite trimethylaminopropionic acid (TMAVA) and clarified its mechanism of aggravating fatty liver and cardiac hypertrophy, and has developed a mass spectrometry detection method for the important gut microbiota active metabolite trimethylamine (TMAO) and discovered its new pathophysiological functions in stroke and pulmonary arterial hypertension. Through metabolic intervention and metabolic-related regulation, we have proposed new ideas and insights for the treatment of cardiovascular and cerebrovascular diseases and metabolic-related diseases.
3. Conduct translational medical research on cardiovascular and cerebrovascular diagnosis and treatment using new materials and technologies. Use new technologies to develop diagnostic and treatment methods for cardiovascular and cerebrovascular diseases, including using bio-nanomaterials carrying imaging groups or pharmacological components to achieve non-invasive imaging or targeted treatment of diseases such as arterial plaques, using molecular probes and structure prediction to develop drugs to correct metabolic disorders related to diseases, optimizing clinical mass spectrometry detection methods to develop a multi-index comprehensive early warning system for cardiovascular and cerebrovascular diseases, etc., to help cardiovascular and cerebrovascular diagnosis and treatment continue to develop on a new track.
